Introducing DCITE: The third wave of T cell engineering

Rather than engineering T cells directly, the next breakthrough in cancer immunotherapy will come from teaching the body’s own dendritic cells to create more powerful, targeted immune responses. Read on for more on how Corner Therapeutics’ DCITE platform is pioneering this third wave of T cell engineering.

A brief history of T cell engineering
Since the discovery of T cells in the 1960s, scientists have been working to harness their disease-fighting power to treat cancer.

In the late 1980s and 1990s, researchers first discovered they could extract T cells from a patient’s body, modify them in the lab and then reintroduce them to the patient. These early efforts at ex vivo T cell engineering later led to the development of CAR-T cells. These modified T cells are better able to recognize specific cancer antigens, giving the body new tools to fight back.

The first FDA-approved CAR-T therapy hit the market in 2017, representing a breakthrough in cancer treatment. While it has proved effective for certain blood cancers like leukemia and lymphoma, it has also faced challenges. CAR-T patients typically require “bridging chemotherapy” preconditioning prior to treatment, which brings with it risks and toxicities. Slow turnaround time and high costs for a patient’s personalized treatment have resulted in limited demand. Furthermore, CAR-T has not been successful for treating solid tumors, which make up approximately 90% of cancers. Despite initial excitement around the first generation of CAR-T therapy, it is clear that significant improvements are required.

The Current State of Play: Early in vivo Exploration
We’re now seeing a second wave of T cell engineering efforts in oncology, with large pharma companies like AbbVie and AstraZeneca acquiring new players focusing on in vivo CAR-T approaches that aim to modify T cells directly within a patient’s body. These approaches use a variety of methods such as lipid nanoparticles (LNPs), mRNA, or viral vectors to directly deliver genetic instructions to targeted T cells, without the need for extraction and reintroduction.

In vivo CAR-T therapies are still in the early stages of development but have generated a buzz in the industry as pharma companies look for ways to extend the benefits of CAR-T therapies to more patients.

The Next Frontier: DCITE

DCs teach the body to generate powerful T cells that are potent, durable, and tumor-specific.

However, a third wave of T cell engineering is poised to transform the industry: dendritic cell-initiated T cell engineering (DCITE).

At Corner Therapeutics, we are developing two in vivo therapeutic platforms that work by stimulating nature’s T cell engineers: dendritic cells (DCs). DCs teach the body to generate powerful T cells that are potent, durable, and tumor-specific. They do so by leveraging the broad “natural intelligence” of the immune system. DCITE-induced T cells can be generated against any type of tumor or infectious disease antigen to protect the body from cancers and pathogens.

DCITE’s LNP-based, mRNA-compatible platforms deliver the precise genetic instructions needed to induce durable, highly targeted immune responses to common cancer antigens, and to a patient’s broad portfolio of tumor antigens if administered intratumorally. By targeting the cells our bodies naturally use to generate T cells, the DCITE platform overcomes current limitations of both traditional ex vivo CAR-T and the latest in vivo approaches.

Ex vivo CAR-T Limitations:

  • Weak performance in solid tumors: Engineered T cells struggle to enter the microenvironment of solid tumors, rendering them ineffective.
  • Long needle-to-needle turnaround time: Extracting, engineering and reintroducing T cells to the body takes weeks. Time is of the essence for cancer patients, and many can’t afford to wait for treatment.
  • High costs: The complex nature of CAR-T therapies makes them incredibly expensive to manufacture and administer. Treatment for a single patient can cost between $500,000 to $1 million, making it inaccessible for many.
  • Dangerous side effects: CAR-T can cause cytokine release syndrome (CRS) and Immune Effector Cell-Associated Neurotoxicity Syndrome (ICANS), leading to a wide range of symptoms including fever, chills, nausea, vomiting, seizures, paralysis and organ damage. Preconditioning chemotherapy also adds risks and toxicities to CAR-T approaches.

In vivo CAR-T limitations:

  • Weak performance in solid tumors: As with ex vivo approaches, in vivo engineered T cells also struggle to enter the microenvironment of solid tumors.
  • Dangerous side effects: in vivo CAR-T can also cause CRS and ICANS.
  • Manufacturing complexity: Though initially meant to simplify the ex vivo manufacturing process, in vivo CAR-T approaches present new challenges due to complex formulations.

DCITE Improvements:

  • Solid Tumor Activation: Recruits and activates T cells directly within the tumor microenvironment if administered intratumorally.
  • Durability: Corner’s platform emphasizes T cell quality and persistence which are critical for long-term efficacy.
  • Non-Systemic Delivery: Intramuscular or intratumoral administration avoids systemic toxicity, reduces manufacturing complexity, and supports repeat dosing—translating to lower cost and greater patient accessibility.
  • Off-the-Shelf Simplicity: LNP-based injectable product—no need for complex viral vectors or systemic delivery.
  • Flexible Modularity: DCITE-STING and DCITE-Inflammasome variants allow tailored T cell responses based on clinical context.

By targeting the cells our bodies naturally use to generate T cells, the DCITE platform overcomes current limitations of both traditional ex vivo CAR-T and the latest in vivo approaches.

Building the cancer treatments of the future
Preclinical murine studies and studies of human tumors demonstrate that our DCITE platform outperformed clinical checkpoint inhibitors and conventional DC stimuli like Alum. As we advance toward clinical trials, Corner Therapeutics is positioned to redefine cancer immunotherapy by making personalized, durable treatment accessible to the patients who need it most. We’re actively seeking strategic partnerships to accelerate development and bring this transformative approach to market.
